Experiment 2: Determination of crude fibre content in foods

Step1: Add 2 g pre-dried powdered sample in that crucible and record as Initial weight.

place crucible add sample ON TARE
0.0 gms

Step2: Measure 200 ml of 0.128 M Sulphuric acid and pour in a conical flask

Measure 100ml

Step3: Add 2 gm sample into the conical flask

Pour sample

Step4: Put the sample flask on hot plate and boil for 30 mins

Turn on Place flask
90C

After 30 minutes

Step5: filter the boiled sample solution using filter cloth

Place filter cloth Filter sample Remove sample

Step6: Measure 200 ml of 0.313 M Sodium hydroxideand pour in a conical flask

Measure 100ml

Step7: Mix the sample residue obtained in step 7 to the conical flask having NaOH

Add sample

Step8: Put the sample flask on hot plate and boil for 30 mins

Turn on Place flask
90C

After 30 minutes

Step9: Filter the boiled sample solution using filter cloth

Place filter cloth Filter sample Remove sample

Step10: Collect the fibre in crucible

Collect sample

Step11: Set the temperature of hot air oven to 130C and dry for 2 hrs

Open Lid Place Sample
PT 000C
ST 000C

After 2 hours

Step12: Weigh the sample crucible and record the weight

place crucible ON TARE
0.0 gms

Step13: Put this sample crucible in muffle furnace and set the temperature to 500°C

Open Lid Place Crucible Close Lid Turn On
PT 000C
ST 000C

After 4 hours

Step14: Then take out sample crucible carefully with help of tong and confirm proper ashing of sample

Open Lid Remove Crucible Close Lid

Step15: Weigh the sample crucible and record the weight

place crucible ON TARE
0.0 gms

Result & calculation

formula

Weight of sample (Ws)= 2 gm

Weight of crucible with fibre (Wf) = 22.15 gm

Weight of crucible with ash (Wa) = 22.08 gm

Crude fiber(%) = 22.15-22.08/2 = 0.035
