Experiment 1: Determination of Total and Acid insoluble ash content in foods

Step1: Add 2 g pre-dried powdered sample in that crucible and record as Initial weight.

place crucible add sample ON TARE
0.0 gms

Step2: Put this sample crucible in muffle furnace and set the temperature to 500°C

Open Lid Place Crucible Close Lid Turn On
PT 000C
ST 000C

Switch off the muffle furnace after 5 hrs and wait for 1 hr.

Step3: Then take out sample crucible carefully with help of tong and confirm proper ashing of sample

Open Lid Remove Crucible Close Lid

Step4: Take weight of sample crucible and recorded as final weight of sample

place crucible ON TARE
0.0 gms

Total Ash Content will be calculated using the following equation

formula

Weight of Crucible (gms) = 10

Weight of Initial Sample (gms) = 2

Weight of Final Sample (gms) = 11.5 - 10 = 1.5

Total Ash(%) = 1.5 / 2 * 100 = 75%

Step5: Take the ash in a beaker and add 25 mL of hydrochloric acid to it.

Put ash in beaker Pour hydrochloric acid

Step6: Heat the sample beaker for 5 minutes using hot plate

Turn On Place Beaker Remove Beaker

Step7: Filter it carefully using ashless filter-paper

Place filter paper Place Funnel Pour Acid

Step8: Put the sample crucible in muffle furnace and set the temperature to 500°C

Open Lid Place Crucible Close Lid Turn On
PT 000C
ST 000C

Step9: Then take out sample crucible carefully

Open Lid Remove Crucible Close Lid

Step10: Take weight of sample crucible and recorded as final weight of sample

place crucible ON TARE
0.0 gms

Total Insoluble Ash Content will be calculated using the following equation

formula

Weight of Crucible (gms) = 10

Weight of Initial Sample (gms) = 2

Weight of Final Sample (gms) = 11 - 10 = 1

Total Insoluble Ash(%) = 1 / 2 * 100 = 50%
